Luciferase Reporter System for Hippo Pathway Readout.

This system use Gal4-TEAD4 fusion protein as the sensor of the activation of Hippo kinase cascade. The transcription factor YAP works together with Gal4-TEAD4 to activate luciferase expression from pGBD-Hyg-Luc vector when transfected into mammalian cells. Activated Hippo kinase cascade results in the phosphorylation of YAP and its cytoplasmic translocation. Less YAP in the nucleus, in turn, cause reduced luciferase expression while inhibition of Hippo kinase cascade results in elevated luciferase activity. This system is very useful for studying the effects of the various components and for screening inhibitors of the Hippo pathway.

Although the system can be used to study Hippo pathway by transient transfection assays, it is especially useful for making stable cell lines for drug screening because both vectors (pGal4-TEAD4 and pBGD-Hyg-Luc) have different antibiotics resistance marker (G418 & Hygromycin, respectively).
